Overview

Calycosin is an O-methylated isoflavone and a typical phytoestrogen extracted primarily from the root of Astragalus membranaceus. It exhibits a wide range of pharmacological activities including anticancer, anti-inflammatory, anti-osteoporosis, neuroprotective, and hepatoprotective effects. Calycosin acts as an estrogen receptor partial agonist and modulates multiple signaling pathways such as PI3K/Akt/mTOR, WDR7-7-GPR30, Rab27B-β-catenin-VEGF among others. It has demonstrated the ability to suppress tumor cell proliferation, induce apoptosis in cancer cells (notably breast cancer), inhibit metastasis in various cancers by regulating ERβ receptor activity and enhance the efficacy of certain chemotherapeutic drugs. Calycosin also shows antioxidant properties and can protect cardiac muscle cells from damage via activation of protective pathways like transient receptor potential paradigm 6. Its pharmacokinetics and toxicology have been studied with suggestions for future clinical trials to optimize dosing and explore synergistic drug combinations[1][2][3][4][5].
