CAM-FAP-Ac-225 is an investigational targeted alpha therapy (TAT) radiopharmaceutical being developed by Precirix for the treatment of fibroblast activation protein (FAP)-positive solid tumors. The drug consists of a single-domain antibody (sdAb) vector that specifically targets FAP, a protein highly expressed on tumor-associated fibroblasts within the stroma of various epithelial-derived cancers, including pancreatic ductal adenocarcinoma (PDAC) and gastric cancer. This targeting vector is conjugated to Actinium-225, a potent alpha-emitting radioisotope. Upon binding to FAP in the tumor microenvironment, CAM-FAP-Ac-225 delivers high-energy alpha particles that induce localized cytotoxicity through double-strand DNA breaks. This mechanism allows for potent anti-tumor activity with minimal off-target effects due to the short range and high linear energy transfer of alpha radiation.
