CAM-H2 is a targeted radionuclide therapeutic drug (TRNT) designed for the treatment of HER2-expressing cancers, including metastatic breast cancer and gastric/gastroesophageal cancer. It consists of a single-domain antibody fragment (sdAb), derived from camelid antibodies, that specifically binds to the human epidermal growth factor receptor 2 (HER2). This sdAb is covalently linked to the radioactive isotope iodine-131 via the SGMIB linker. Upon administration, CAM-H2 delivers beta radiation directly to HER2-positive tumor cells while minimizing exposure to healthy tissues. The drug has demonstrated high specificity and retention in HER2+ tumors in imaging studies and has shown a favorable safety profile in phase I clinical trials. It is currently being evaluated in phase I/II clinical studies for patients who have relapsed or are refractory to existing anti-HER2 therapies[1][4][5][6][8].
