Overview

camB7-H3 CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat solid tumors. This biologic therapy utilizes dromedary camel-derived nanobodies (VHH) as the antigen-binding domain, which specifically target the IgC domain of the B7-H3 (CD276) transmembrane protein. B7-H3 is a member of the B7 family of immune checkpoint proteins and is frequently overexpressed in various cancers, harboring two distinct epitope motifs in its ectodomain: IgC and IgV. While most existing B7-H3-targeted agents bind to the IgV domain, camB7-H3 CAR T cells specifically recognize the IgC domain. This specific epitope targeting is intended to optimize the CAR's antitumor activity and potentially reduce off-target effects. Preclinical studies have demonstrated that these CAR T cells possess potent activity against large, established solid tumors in mouse models, highlighting their potential as a therapeutic candidate for human solid tumor treatment.
