Overview

HuMax-TAC (camidanlumab) is a high-affinity, fully human IgG1 monoclonal antibody developed by Genmab that targets the alpha chain of the interleukin-2 receptor (IL-2Rα), also known as CD25. CD25 is a component of the high-affinity IL-2 receptor and is overexpressed on various hematological malignancies and regulatory T cells (Tregs), while showing limited expression on normal resting tissues. By binding to CD25, HuMax-TAC blocks the interaction between IL-2 and its receptor, thereby inhibiting IL-2-dependent proliferation of T cells. Additionally, as an IgG1 antibody, it can induce antibody-dependent cellular cytotoxicity (ADCC) against CD25-positive cells. While initially explored for autoimmune diseases, transplant rejection, and inflammation, its primary clinical significance evolved as the targeting moiety for the antibody-drug conjugate camidanlumab tesirine (ADCT-301).
