Overview

Camidanlumab tesirine is an investigational antibody-drug conjugate (ADC) designed for the treatment of hematological malignancies, particularly relapsed or refractory classical Hodgkin lymphoma and acute myeloid leukemia. It consists of a human IgG1 monoclonal antibody targeting CD25 (the alpha chain of the interleukin 2 receptor, IL2RA), which is overexpressed on activated T and B cells in various leukemias and lymphomas. The antibody is conjugated via a cleavable linker to a highly potent pyrrolobenzodiazepine (PBD) dimer toxin payload called tesirine. Upon binding to CD25-expressing cells, the ADC is internalized, releasing the PBD payload intracellularly where it crosslinks DNA minor grooves, leading to cell death by blocking cell division. This mechanism also allows for bystander killing of neighboring tumor cells and may induce immunogenic cell death, potentially enhancing anti-tumor immune responses[1][3][4][8].
