Camizestrant is an orally available, next-generation selective estrogen receptor degrader (SERD) and complete estrogen receptor antagonist developed for the treatment of hormone receptor-positive (HR+), HER2-negative breast cancer. It binds to the estrogen receptor (ER), induces a conformational change, and promotes proteasome-mediated degradation of the ER, thereby blocking ER-mediated signaling and inhibiting proliferation of ER-expressing tumor cells. Camizestrant is effective against both wild-type and mutant forms of ERα, with no evidence of agonist activity in uterine models. It is administered orally and has demonstrated anti-cancer activity in preclinical models as well as clinical trials, including improved progression-free survival compared to fulvestrant in patients with advanced or metastatic breast cancer.
