Camizestrant + atirmociclib is an investigational combination therapy developed by AstraZeneca for the treatment of estrogen receptor-positive (ER+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer. Camizestrant (AZD9833) is a next-generation, potent, oral selective estrogen receptor degrader (SERD) that functions by binding to, antagonizing, and inducing the degradation of the estrogen receptor alpha (ERα), including common ESR1 mutations. Atirmociclib (AZD9829) is a highly selective cyclin-dependent kinase 4 (CDK4) inhibitor. By specifically targeting CDK4 over CDK6, atirmociclib aims to reduce the hematological toxicities, such as neutropenia, typically associated with dual CDK4/6 inhibitors while maintaining efficacy in inhibiting cell cycle progression. This combination is being evaluated in clinical trials, such as SERENA-1b, to improve outcomes for patients who have previously received CDK4/6 inhibitor therapy.
