Camptothesome is a novel nanoparticle-based drug delivery system designed to encapsulate and deliver camptothecin or its derivatives for cancer therapy. It is not itself a small molecule drug, but rather a nanovesicle formulation engineered to improve the pharmacokinetics, stability, and tumor-targeting of camptothecin-class topoisomerase I inhibitors. The core mechanism of action derives from its payload—camptothecin or related analogues—which inhibit DNA topoisomerase I by stabilizing the cleavable complex between the enzyme and DNA, leading to S-phase-specific cytotoxicity in rapidly dividing tumor cells. The vesicular structure of Camptothesome enhances solubility and bioavailability while reducing systemic toxicity compared to free camptothecin. This platform is under preclinical or early clinical development for various solid tumors.
