This is a combination regimen consisting of five drugs: - **Camrelizumab** is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to enhance anti-tumor immune responses. - **Fluorouracil (5-FU)** is an antimetabolite chemotherapy agent that inhibits thymidylate synthase, disrupting DNA and RNA synthesis in rapidly dividing cells. - **Leucovorin (folinic acid)** enhances the binding of 5-FU to its target enzyme, increasing its cytotoxic effect. - **Oxaliplatin** is a platinum-based chemotherapeutic that causes DNA cross-linking and apoptosis in cancer cells. - **Docetaxel** is a taxane-class chemotherapeutic that stabilizes microtubules, inhibiting cell division. The four-drug combination of fluorouracil, leucovorin, oxaliplatin, and docetaxel—known as FLOT—is established for perioperative treatment of gastric and gastroesophageal junction adenocarcinoma[2][3][4][5][6][7]. The addition of camrelizumab introduces immunotherapy to this regimen. This five-drug combination aims to combine cytotoxic chemotherapy with immune checkpoint blockade for enhanced anti-tumor efficacy. Camrelizumab was developed by Jiangsu Hengrui Medicine.
