Overview

This combination therapy is a multi-modal treatment regimen being investigated for patients with unresectable hepatocellular carcinoma (HCC), specifically those who have failed transcatheter arterial chemoembolization (TACE). The regimen integrates three distinct therapeutic approaches: immunotherapy with **camrelizumab** (a PD-1 inhibitor), targeted therapy with a tyrosine kinase inhibitor (TKI) such as **lenvatinib** or **regorafenib**, and localized chemotherapy via **hepatic artery infusion chemotherapy (HAIC)**. Camrelizumab works by blocking the PD-1/PD-L1 pathway to restore T-cell anti-tumor activity. The TKIs inhibit multiple pathways involved in angiogenesis and tumor cell proliferation, including VEGFR and FGFR. HAIC allows for the delivery of high concentrations of cytotoxic chemotherapy directly into the tumor-supplying artery, maximizing local efficacy while minimizing systemic toxicity. This synergistic combination aims to improve objective response rates and survival in advanced liver cancer patients.
