Overview

This is a combination therapy consisting of three agents: - **Camrelizumab** is a fully humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated antitumor responses. - **Nab-paclitaxel** is an albumin-bound formulation of paclitaxel, a microtubule inhibitor that disrupts cell division and induces apoptosis in cancer cells. - **Famitinib** is a small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptor 2 (VEGFR2), platelet-derived growth factor receptor (PDGFR), and c-kit, thereby inhibiting angiogenesis and tumor proliferation. The triplet regimen has been investigated as first-line treatment for patients with unresectable, locally advanced, or metastatic immunomodulatory triple-negative breast cancer (TNBC). Clinical studies have shown high objective response rates (~81%) and manageable toxicity profiles in this population[1][3][6]. The rationale for the combination includes synergistic effects between immune checkpoint inhibition, antiangiogenic therapy, and cytotoxic chemotherapy[8].
