Overview

This is a combination therapy consisting of camrelizumab, a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), and pyrotinib, an irreversible pan-HER tyrosine kinase inhibitor. The combination is being investigated primarily for the first-line treatment of advanced or metastatic HER2-positive gastric and gastroesophageal junction (G/GEJ) adenocarcinoma. Camrelizumab acts as an immune checkpoint inhibitor by blocking PD-1, thereby enhancing T-cell mediated anti-tumor immunity. Pyrotinib inhibits multiple members of the human epidermal growth factor receptor (HER) family, including HER2, leading to inhibition of tumor cell proliferation and survival. Clinical studies have shown promising efficacy with this combination in HER2-positive G/GEJ cancer when used with chemotherapy[1][2][3].
