CAN-225-IR700 is a canine-specific antibody-photo-absorber conjugate (APC) designed for near-infrared photoimmunotherapy (NIR-PIT). It consists of can225IgG, a recombinant caninized monoclonal antibody engineered from the antigen-binding site of cetuximab, which is conjugated to the silica-phthalocyanine photo-absorber dye IRDye 700DX (IR700). The drug specifically targets the epidermal growth factor receptor (EGFR), which is highly expressed in various canine cancers, including invasive transitional cell carcinoma (TCC) of the bladder. Upon binding to the target cells and subsequent exposure to near-infrared light (approximately 690 nm), the conjugate induces rapid, selective necrotic cell death. This modality is intended to provide a highly targeted treatment with minimal off-target effects and has been shown to stimulate an immunogenic response. It is currently being evaluated in preclinical models as a prelude to clinical trials in pet dogs with spontaneous EGFR-expressing cancers.
