CAN016 is a novel, dual-payload antibody-drug conjugate (ADC) being developed by CanWell Pharma. It is designed to target Human Epidermal Growth Factor Receptor 2 (HER2) and incorporates two distinct cytotoxic agents within a single antibody construct. This synergistic dual-payload approach is intended to overcome resistance mechanisms that limit the efficacy of traditional single-payload ADCs. Preclinical studies in cell-line-derived (CDX) and patient-derived xenograft (PDX) models have shown potent anti-tumor activity, including in models resistant to currently approved HER2-targeted ADCs. The FDA has cleared the IND application for CAN016, supporting its evaluation in a Phase I clinical trial for patients with advanced solid tumors who have progressed on prior ADC therapies.
