CAN04 + gemcitabine + nab-paclitaxel is a three-drug combination regimen for the treatment of advanced or metastatic pancreatic cancer. CAN04 (nadunolimab) is a fully humanized monoclonal IgG1 antibody targeting Interleukin-1 Receptor Accessory Protein (IL1RAP), blocking IL-1α and IL-1β signaling and triggering antibody-dependent cellular cytotoxicity (ADCC). Gemcitabine is a nucleoside analog antineoplastic agent inhibiting DNA synthesis, and nab-paclitaxel (albumin-bound paclitaxel) is a microtubule inhibitor delivered via nanoparticle-formulated albumin for improved tumor uptake. This regimen is being investigated for increasing efficacy and clinical benefit beyond the standard gemcitabine + nab-paclitaxel combination.
