Overview

CAN1012 is a selective and potent small molecule agonist of Toll-like receptor 7 (TLR7), developed for intratumoral administration in the treatment of advanced solid tumors. By binding to and activating TLR7, CAN1012 stimulates innate immune responses—particularly the activation of NF-κB pathways and robust production of interferon-alpha (IFN-α) by plasmacytoid dendritic cells. This leads to enhanced infiltration of CD4+ and CD8+ T cells into the tumor microenvironment while reducing immunosuppressive myeloid-derived suppressor cells (MDSCs) and tumor-associated macrophages (TAMs). Preclinical studies have demonstrated significant antitumor activity with low systemic toxicity due to high local exposure after intratumoral injection. Clinical trials are ongoing in patients with advanced or unresectable solid tumors who are not candidates for standard therapy[1][3][6].
