Canakinumab is a fully human monoclonal antibody of the IgG1/κ isotype that specifically targets and neutralizes interleukin-1 beta (IL-1β), a pro-inflammatory cytokine implicated in several autoinflammatory and inflammatory diseases. By binding to IL-1β with high affinity, canakinumab prevents its interaction with the interleukin-1 receptor, thereby inhibiting downstream inflammatory signaling. It does not bind to IL-1α or the IL-1 receptor antagonist. Canakinumab was developed by Novartis and is primarily indicated for the treatment of periodic fever syndromes such as Cryopyrin-Associated Periodic Syndromes (CAPS), Familial Mediterranean Fever (FMF), Tumor Necrosis Factor Receptor Associated Periodic Syndrome (TRAPS), Mevalonate Kinase Deficiency/Hyperimmunoglobulin D Syndrome (MKD/HIDS), Still's disease including adult-onset Still's disease, systemic juvenile idiopathic arthritis (SJIA) in children, and gout flares in adults who are refractory or intolerant to standard therapies[1][2][4][7].
