Overview

Canakinumab + spartalizumab is an investigational combination therapy consisting of two monoclonal antibodies developed by Novartis. Canakinumab is a high-affinity human monoclonal antibody targeting interleukin-1 beta (IL-1β), neutralizing its inflammatory activity and thereby reducing inflammation. Spartalizumab is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to enhance T-cell mediated antitumor responses. This combination has been studied in early-phase clinical trials for various cancers, including metastatic pancreatic cancer and localized clear cell renal cell carcinoma, with the aim of leveraging both anti-inflammatory and immunomodulatory mechanisms to improve antitumor efficacy[2][3][4][5][7].
