Overview

This is a four-drug combination regimen consisting of canakinumab, spartalizumab, nab-paclitaxel, and gemcitabine. Canakinumab (ACZ885, brand name Ilaris) is a high-affinity human monoclonal antibody targeting interleukin-1β (IL-1β), thereby reducing inflammation. Spartalizumab (PDR001) is a monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) receptor on T cells to enhance anti-tumor immune responses. Nab-paclitaxel (brand name Abraxane) is an albumin-bound formulation of paclitaxel that inhibits microtubule disassembly, disrupting cell division. Gemcitabine is a nucleoside analog that interferes with DNA synthesis and induces apoptosis in rapidly dividing cells. This quadruplet regimen has been investigated in phase 1b/2 clinical trials for metastatic pancreatic cancer to determine safety and tolerability; it combines immunotherapy with standard chemotherapy[1][3][4][5][8].
