Canalevia-CA1 is a conditionally approved, prescription antidiarrheal drug for dogs, specifically indicated for the treatment of chemotherapy-induced diarrhea (CID). The active ingredient is crofelemer, a botanical drug substance derived from the red latex of the Croton lechleri tree. Crofelemer is an oligomeric proanthocyanidin mixture primarily composed of catechin and epicatechin monomers. Its mechanism in humans involves inhibition of both cAMP-stimulated cystic fibrosis transmembrane conductance regulator (CFTR) chloride channels and calcium-activated chloride channels (CaCC) at the luminal membrane of enterocytes, reducing chloride ion secretion and normalizing fluid influx into the gastrointestinal tract. While this mechanism is presumed similar in dogs, it has not been fully characterized in canines. Canalevia-CA1 is administered orally as an enteric-coated tablet containing 125 mg crofelemer[2][3][8]. It was developed to address a significant unmet need for managing diarrhea associated with cancer therapy in dogs[3][4].
