Overview

This is a combination chemotherapy regimen consisting of canfosfamide (also known as TLK286), carboplatin, and paclitaxel. Canfosfamide is a glutathione analog prodrug that is activated by glutathione S-transferase P1-1 (GST P1-1), an enzyme often overexpressed in cancer cells. Upon activation, it releases cytotoxic species that induce apoptosis in tumor cells. Carboplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and inhibits DNA synthesis and function. Paclitaxel stabilizes microtubules and prevents their depolymerization during cell division, leading to cell cycle arrest and apoptosis. This combination has been studied as first-line therapy for advanced or metastatic non-small cell lung cancer and has shown activity with an acceptable safety profile[1]. Maintenance therapy with canfosfamide may further improve outcomes.
