Overview

Cannabidiol + clobazam is a **combination regimen of two distinct active small molecules:** cannabidiol (CBD), a cannabinoid derivative, and clobazam, a 1,5-benzodiazepine anticonvulsant. This combination is primarily used as adjunct therapy for treatment-resistant epilepsies, such as Dravet syndrome and Lennox–Gastaut syndrome[1][2][3]. CBD enhances the plasma levels of clobazam and its active metabolite norclobazam by inhibiting CYP3A4 and CYP2C19 metabolic pathways, leading to a pharmacokinetic interaction[1][3]. In addition, these agents appear to have a pharmacodynamic interaction: both modulate γ-aminobutyric acid (GABA) A receptor activity, with a combined effect resulting in enhanced inhibitory neurotransmission, further potentiating anticonvulsant efficacy[1]. The regimen is associated with greater efficacy in seizure reduction than either agent alone but comes with an increased risk of drowsiness and sedation due to elevated clobazam levels[2][3]. This drug combination does not currently appear to have a unique marketed brand name; rather, it refers to the co-prescription of the two individual drugs.
