cannabidiol + DANA is an experimental combination therapy consisting of the non-psychoactive phytocannabinoid cannabidiol (CBD) and the sialidase inhibitor DANA (2,3-didehydro-2,4-dideoxy-N-acetylneuraminic acid). This combination is primarily investigated for its synergistic potential in preventing or treating viral infections, specifically COVID-19, by modulating host cell entry factors. Cannabidiol is thought to downregulate the expression of Angiotensin-converting enzyme 2 (ACE2) and Transmembrane protease serine 2 (TMPRSS2) receptors, potentially through the activation of peroxisome proliferator-activated receptor gamma (PPAR-gamma). DANA, a transition-state analog of sialic acid, acts as a pan-sialidase inhibitor that blocks the activity of neuraminidases (sialidases) involved in modifying cell surface glycans, a process that can facilitate viral attachment and penetration. Research conducted by Pathway Rx and the University of Lethbridge suggests that the combination significantly reduces the protein levels of ACE2 and TMPRSS2 in human oral, airway, and intestinal tissues more effectively than either compound administered alone.
