Overview

Canstatin is an endogenous angiogenesis inhibitor derived from the noncollagenous (NC1) domain of the α2 chain of type IV collagen. It is a 24-kDa fragment of basement membrane collagen that functions as a matrix-derived inhibitor of angiogenesis and tumor growth. Canstatin exerts its anti-tumor effects through multiple mechanisms: it inhibits endothelial cell proliferation, migration, and tube formation; induces apoptosis specifically in endothelial cells through mitochondrial damage and down-regulation of the anti-apoptotic protein FLIP; and acts through interaction with αvβ3 and αvβ5 integrin receptors. The protein triggers mitochondrial apoptotic mechanisms through procaspase-9 cleavage in both endothelial and tumor cells. Canstatin has been produced as a recombinant molecule in *E. coli* and mammalian cell systems for research and potential therapeutic applications. It has demonstrated efficacy in suppressing tumor growth in various cancer xenograft models, including pancreatic cancer, hepatocellular carcinoma, breast cancer, prostate cancer, and glioblastoma, primarily through inhibition of angiogenesis and reduction of tumor microvessel density. Beyond oncology, canstatin has shown cardioprotective effects in myocardial infarction models by preventing adverse cardiac remodeling.
