Cantuzumab ravtansine is an antibody-drug conjugate (ADC) developed for the treatment of cancers. It consists of a humanized monoclonal antibody, cantuzumab (huC242), which targets Tumor-associated carbohydrate antigen CanAg, a glycoform of MUC1 that is overexpressed in various tumors such as those of the pancreas, colon, and biliary tract. The antibody is linked via a hindered disulfide linker to the cytotoxic agent DM4 (ravtansine), a maytansinoid that disrupts microtubule assembly by binding tubulin. Upon binding to CanAg on tumor cells and internalization, the ADC releases DM4 intracellularly, leading to inhibition of cell division and induction of apoptosis. Cantuzumab ravtansine was primarily developed by ImmunoGen for indications including gastric cancer and solid tumors but development has been discontinued[1][2][5][6][7].
