Overview

CAP-IL-12 LNP is an investigational gene therapy developed by Earli, designed to selectively deliver and express Interleukin-12 (IL-12) within the tumor microenvironment. The therapy utilizes DNA nanoplasmids encapsulated in lipid nanoparticles (LNPs) for systemic (intravenous) administration. These nanoplasmids are engineered with synthetic cancer-activated promoters (CAPs) that remain transcriptionally inert in healthy tissues but drive high levels of IL-12 expression upon entering malignant cells. By reprogramming tumor cells to secrete IL-12 locally, the treatment aims to stimulate a robust anti-tumor immune response—characterized by the activation of CD4+ and CD8+ T cells and dendritic cells—while avoiding the severe systemic toxicities typically associated with recombinant IL-12 protein administration. Preclinical studies have demonstrated complete ablation of tumor growth in syngeneic models with minimal systemic cytokine exposure.
