CAP1-6D is a modified peptide vaccine derived from the carcinoembryonic antigen (CEA), specifically designed to enhance immunogenicity compared to the native CEA peptide. Developed by researchers at the University of Chicago, it is typically administered as a combination therapy with the adjuvants Montanide ISA-51 and granulocyte-macrophage colony-stimulating factor (GM-CSF/sargramostim). The vaccine targets CEA (CEACAM5), a tumor-associated antigen frequently overexpressed in various cancers, including pancreatic adenocarcinoma. By presenting a modified epitope (6D substitution), the vaccine aims to stimulate a robust T-cell mediated immune response against CEA-expressing tumor cells. It has been evaluated in Phase I/II clinical trials for patients with locally advanced or surgically resected pancreatic cancer.
