Overview

CAP256V2LS is a broadly neutralizing monoclonal antibody targeting the V2 apex of the HIV-1 envelope glycoprotein gp120. It was engineered from the parental antibody CAP256-VRC26.25, with a specific mutation (K127A) to improve manufacturability and prevent proteolytic clipping without altering its neutralization potency or breadth[6]. CAP256V2LS demonstrates high potency and breadth against HIV-1, particularly Clade C viruses, neutralizing 72% of pseudoviruses tested with a median IC50 of 0.001 μg/mL[5]. The antibody is being developed primarily for HIV prevention and treatment, including as part of combination regimens with other broadly neutralizing antibodies such as VRC07-523LS[4][3]. Clinical trials have shown favorable safety and pharmacokinetic profiles in both healthy volunteers and people living with HIV on antiretroviral therapy[4][6].
