Overview

Capecitabine + aromatase inhibitor is a combination therapy used primarily in the treatment of hormone receptor-positive, HER2-negative metastatic breast cancer. Capecitabine is an oral prodrug that is enzymatically converted to 5-fluorouracil (5-FU) in tumor tissue, where it inhibits DNA synthesis and disrupts RNA function, leading to cell death. Aromatase inhibitors (AIs) are a class of drugs that block the enzyme aromatase, thereby reducing estrogen production and limiting growth stimulation of estrogen receptor-positive breast cancer cells. The combination has demonstrated synergistic antitumor effects by targeting both DNA synthesis and hormonal pathways. Clinical studies have shown improved progression-free survival and overall response rates with this regimen compared to either agent alone, with an acceptable safety profile[1][2][4][5].
