Overview

Capecitabine + cisplatin + sorafenib is a combination chemotherapy regimen investigated primarily for the treatment of advanced gastric cancer and other solid tumors. Capecitabine is an oral prodrug of 5-fluorouracil that inhibits thymidylate synthase and disrupts DNA synthesis in tumor cells. Cisplatin is a platinum-based chemotherapeutic agent that forms DNA crosslinks leading to apoptosis. Sorafenib is a small molecule multikinase inhibitor targeting RAF kinases as well as vascular endothelial growth factor receptors (VEGFR) and platelet-derived growth factor receptor (PDGFR), thereby inhibiting tumor cell proliferation and angiogenesis. This combination has been studied in phase II clinical trials for metastatic gastric cancer but did not demonstrate improved efficacy over standard regimens[1]. The regimen has also been explored in hepatocellular carcinoma[2].
