Overview

Capecitabine + gemcitabine + oxaliplatin is a triple-drug chemotherapy regimen used primarily in the treatment of advanced or metastatic pancreatic cancer and biliary tract cancers. This combination brings together three cytotoxic agents with distinct mechanisms of action: - Capecitabine is an oral prodrug that is converted to 5-fluorouracil (5-FU) preferentially in tumor tissue, where it inhibits thymidylate synthase and disrupts DNA synthesis. - Gemcitabine is a nucleoside analog that incorporates into DNA during replication, causing chain termination and apoptosis. - Oxaliplatin is a platinum-based agent that forms cross-links within DNA strands, inhibiting replication and transcription. The regimen has been studied under names such as GEMOXEL. Clinical trials have shown this combination to be feasible with manageable toxicity profiles and promising efficacy as first-line therapy for advanced pancreatic adenocarcinoma[1][2][3][5]. It has also been explored for use in advanced biliary tract cancers[5].
