XELOXGEM is a combination chemotherapy regimen consisting of gemcitabine, capecitabine, and oxaliplatin. This triplet therapy was evaluated in a multicenter Phase I/II trial led by researchers at Yonsei University's Cancer Metastasis Research Center as a second-line treatment for patients with advanced or metastatic colorectal cancer who had previously failed irinotecan-based therapy. The regimen adds gemcitabine, a nucleoside analog that inhibits DNA synthesis, to the standard XELOX backbone (capecitabine and oxaliplatin). Capecitabine acts as a prodrug of 5-fluorouracil to inhibit thymidylate synthase, while oxaliplatin is a platinum-based agent that forms DNA adducts, collectively providing a multi-targeted approach to disrupting tumor cell division and inducing apoptosis.
