capecitabine + irinotecan + simvastatin

capecitabine + irinotecan + simvastatin is a combination chemotherapy regimen that was evaluated for the treatment of previously treated metastatic colorectal cancer. The regimen combines the standard XELIRI chemotherapy (capecitabine and irinotecan) with simvastatin, an HMG-CoA reductase inhibitor. The rationale for adding simvastatin was based on preclinical evidence suggesting that statins may inhibit tumor cell growth, induce apoptosis, and enhance the effects of chemotherapy through pleiotropic effects on the mevalonate pathway. A randomized Phase III trial conducted in South Korea compared XELIRI/FOLFIRI plus simvastatin to XELIRI/FOLFIRI plus placebo. The study concluded that the addition of 40 mg of simvastatin did not significantly improve progression-free survival (PFS) or overall survival (OS) in this patient population.
