Capecitabine + mitomycin c is a combination chemotherapy regimen used primarily in the treatment of gastrointestinal cancers such as metastatic colorectal cancer and anal cancer. Capecitabine is an orally administered prodrug that is converted to 5-fluorouracil (5-FU) within tumor cells, where it acts as an antimetabolite inhibiting DNA synthesis and slowing tumor growth. Mitomycin C is an antibiotic with antitumor properties that inhibits DNA replication and transcription by cross-linking DNA strands. The combination may have synergistic effects because mitomycin c upregulates thymidine phosphorylase, enhancing the activation of capecitabine to 5-FU within tumors. This regimen can be used beyond standard lines of therapy for metastatic colorectal cancer and in conjunction with radiotherapy for anal cancer[1][2][3][4].
