Capecitabine + nintedanib is an investigational oral combination therapy evaluated primarily for refractory metastatic colorectal cancer. Capecitabine is a prodrug of 5-fluorouracil, functioning as an antimetabolite that inhibits DNA synthesis in rapidly dividing tumor cells. Nintedanib is a small molecule tyrosine kinase inhibitor targeting multiple angiokinase receptors, including vascular endothelial growth factor receptor (VEGFR), platelet-derived growth factor receptor (PDGFR), and fibroblast growth factor receptor (FGFR). This dual approach aims to inhibit tumor cell proliferation and angiogenesis. Clinical studies have shown the combination to be well tolerated with promising efficacy compared to other therapies in heavily pretreated colorectal cancer patients[1][4][5].
