This experimental combination regimen is being evaluated as a neoadjuvant therapy for patients with high-risk, mid-to-low localized rectal cancer. The regimen consists of short-course radiotherapy (SCRT) followed by CapeOX chemotherapy (a combination of capecitabine and oxaliplatin) integrated with toripalimab, a PD-1 inhibitor, and surufatinib, a small molecule tyrosine kinase inhibitor targeting VEGFR and FGFR. This multi-modal approach aims to improve pathological complete response (pCR) rates by combining cytotoxic chemotherapy, radiation, immune checkpoint blockade, and anti-angiogenic therapy. The study is sponsored by The First Hospital of Jilin University and explores whether the addition of surufatinib to the chemo-immunotherapy backbone enhances efficacy in high-risk locally advanced rectal cancer (LARC).
