Overview

This is a three-drug oncology regimen combining the fluoropyrimidine prodrug capecitabine, the platinum chemotherapy oxaliplatin, and the HER2-targeted monoclonal antibody trastuzumab. Capecitabine is converted to 5‑fluorouracil in tumors to inhibit thymidylate synthase and DNA synthesis; oxaliplatin forms DNA crosslinks that block replication and repair; trastuzumab binds HER2 (ERBB2) to inhibit receptor signaling and mediate antibody-dependent cellular cytotoxicity. The combination is used for HER2‑positive gastric or gastro‑oesophageal junction cancers and has been studied as trastuzumab plus XELOX (capecitabine + oxaliplatin) with high response rates in HER2‑positive advanced gastric cancer.[1][4][6]
