Overview

This is a combination therapy consisting of three separate medications used together to treat HER2-positive cancers. Tucatinib is a potent, highly selective HER2-targeted tyrosine kinase inhibitor that inhibits the phosphorylation of both HER2 and HER3 proteins, which in turn inhibits downstream activation of MAPK and AKT signaling pathways and cell proliferation[7]. Trastuzumab is a type of targeted cancer drug called a monoclonal antibody that targets HER2 receptors[1]. Capecitabine is a type of chemotherapy called an anti-metabolite. The body changes capecitabine into fluorouracil, which stops cells from making and repairing DNA[1].
