Overview

Capivasertib is an orally administered small molecule kinase inhibitor that targets all three isoforms of the serine/threonine kinase AKT (AKT1, AKT2, and AKT3). It is used in combination with fulvestrant to treat hormone receptor–positive (HR+), human epidermal growth factor receptor 2–negative (HER2-) locally advanced or metastatic breast cancer in adults whose tumors have one or more PIK3CA/AKT1/PTEN alterations and have progressed on at least one endocrine-based regimen. By inhibiting AKT activity, capivasertib blocks a key component of the PI3K/AKT signaling pathway involved in cell proliferation, survival, and metabolism. This inhibition slows or stops tumor growth by interfering with cancer-promoting signals. Capivasertib was developed by AstraZeneca and is currently marketed under the brand name Truqap.
