**capivasertib + abemaciclib + fulvestrant** is an investigational combination therapy comprising three agents: - **Capivasertib**: a small molecule inhibitor of AKT, a kinase in the PI3K/AKT signaling pathway, which is frequently dysregulated in breast cancer and contributes to cell growth, survival, and endocrine resistance. - **Abemaciclib**: a small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which block cell cycle progression from G1 to S phase, inhibiting tumor cell proliferation. - **Fulvestrant**: a selective estrogen receptor degrader (SERD), which binds, blocks, and induces degradation of the estrogen receptor, preventing estrogen-driven cancer cell growth. This triplet regimen is being studied in hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) locally advanced, unresectable or metastatic breast cancer, including patients with prior resistance to endocrine therapy and/or CDK4/6 inhibitors. The combination targets multiple complementary pathways believed to drive breast cancer progression and therapeutic resistance[1][5].
