Overview

Capmatinib + trametinib is an investigational combination therapy consisting of two small molecule inhibitors: capmatinib, a selective MET tyrosine kinase inhibitor, and trametinib, a MEK inhibitor. This combination is being studied primarily for the treatment of non-small cell lung cancer (NSCLC) with MET exon 14 skipping mutations, MET amplification, or MET fusion. The rationale for this combination arises from evidence that resistance to single-agent MET inhibition can develop through hyperactivation of the MAPK pathway; co-inhibition with a MEK inhibitor like trametinib may overcome this resistance mechanism. Capmatinib targets aberrant signaling driven by alterations in the MET gene, while trametinib inhibits downstream MAPK/ERK signaling by targeting MEK1/2 kinases[1][2][7].
