Overview

Capmatinib + nazartinib is an investigational oral combination therapy for advanced non-small cell lung cancer (NSCLC) with EGFR mutations, particularly in patients who have developed resistance to prior EGFR tyrosine kinase inhibitor (TKI) treatments. Capmatinib is a selective MET kinase inhibitor, while nazartinib is a third-generation EGFR TKI that targets activating and resistance mutations such as T790M. The combination aims to overcome acquired resistance mechanisms involving both MET amplification and secondary EGFR mutations. In phase 1b/2 clinical trials, this regimen demonstrated antitumor activity in patients with EGFR-mutated NSCLC resistant to previous TKIs, with an acceptable safety profile. The most common adverse events included peripheral edema, nausea, diarrhea, and rash[1][2][3][4][5].
