**CAPN2-MSN** is a preclinical, calpain-2-responsive mesoporous silica nanoparticle delivery system developed for targeted treatment of pancreatic ductal adenocarcinoma. The nanoparticle is loaded with **paclitaxel** and sealed using a CAPN2-cleavable protease-responsive molecular gate; after cellular uptake by pancreatic cancer cells, intracellular calpain-2 cleaves the gate and releases paclitaxel. In pancreatic cancer cell models and two mouse models, paclitaxel-loaded CAPN2-MSN reduced tumor growth at least as effectively as free paclitaxel while reducing leukopenia and limiting toxicity to healthy cells and organs.
