Overview
The Caprin-1 ISAC is an immune-stimulating antibody conjugate (ISAC) developed through a collaboration between Bolt Biotherapeutics and Toray Industries. Utilizing Bolt's proprietary Boltbody™ platform, the drug consists of Toray's TRK-950, a monoclonal antibody targeting the tumor-associated antigen Caprin-1, conjugated to a TLR7/8 agonist payload via a non-cleavable linker. Caprin-1 is highly expressed on the cell surface of various solid tumors but has limited expression in normal tissues. By targeting Caprin-1, the ISAC delivers the immune stimulant directly to the tumor microenvironment, where it activates myeloid cells—such as macrophages and dendritic cells—to induce a potent, localized anti-tumor immune response and potentially generate long-term immunological memory.
