Overview

**Capsazepine** is a synthetic small molecule developed as a *competitive antagonist* of the **transient receptor potential vanilloid 1 (TRPV1) receptor**, which is a non-selective cation channel involved in pain perception, temperature regulation, and inflammation[1][4][7][9][10][13][16]. Its mechanism of action involves blocking TRPV1 activation, thus inhibiting the painful sensation typically triggered by capsaicin, the active compound in chili peppers[1][7]. Capsazepine also exhibits antagonistic activity at TRPA1 and can inhibit other channels including TRPM8, voltage-gated calcium channels, and nicotinic acetylcholine receptors[7][22]. Beyond its use in research as a TRPV1 antagonist, capsazepine has demonstrated anti-inflammatory and anticancer activities in preclinical studies—suppressing proliferation and inducing apoptosis in various cancer cell lines (prostate, breast, colorectal, oral, and osteosarcoma)[1][4][6]. The drug modulates the JAK/STAT pathway, induces ROS and CHOP (causing programmed cell death), and downregulates NF-κB, contributing to its pleiotropic pharmacological effects[1][4][6]. Discovered and first characterized by Sandoz (now Novartis), capsazepine is primarily used as a biochemical tool and for preclinical investigations due to poor pharmacokinetics limiting its clinical development[4][7][13].
