CAR-ABCA1 mRNA is an experimental mRNA-based therapeutic designed to reprogram macrophages for the treatment of atherosclerosis. Delivered via Ly6C-targeted lipid nanoparticles (Ly6C/LNPs), the mRNA encodes a dual-functional fusion construct consisting of a chimeric antigen receptor (CAR PS-FcRγ) and the ATP-binding cassette transporter A1 (ABCA1). The CAR component is engineered to enhance efferocytosis (the clearance of apoptotic cells) by binding to phosphatidylserine, while ABCA1 promotes cholesterol efflux, thereby preventing the formation of lipid-laden foam cells. By simultaneously addressing defective clearance and metabolic imbalances within atherosclerotic plaques, this therapy aims to stabilize lesions, reduce inflammation, and increase fibrous cap thickness.
