Overview

CAR-CLL1 is an investigational chimeric antigen receptor (CAR) T-cell therapy targeting C-type lectin-like molecule-1 (CLL1, also known as CLEC12A). CLL1 is a type II transmembrane glycoprotein highly expressed on the surface of acute myeloid leukemia (AML) blasts and leukemic stem cells (LSCs), while being largely absent on normal hematopoietic stem cells (HSCs). This expression profile makes CLL1 an attractive target for reducing off-tumor toxicity and preventing disease relapse. The therapy involves genetically engineering T cells (either autologous or donor-derived) to express a CAR that specifically recognizes the CLL1 antigen. Upon infusion, these CAR-T cells bind to CLL1-positive leukemia cells, leading to T-cell activation, cytokine release, and targeted cell lysis. CAR-CLL1 is primarily being evaluated for the treatment of relapsed or refractory AML, often as a bridge to allogeneic hematopoietic stem cell transplantation.
