Overview

CAR-DNT (Chimeric Antigen Receptor Double-Negative T cells) is an allogeneic adoptive cellular therapy platform that utilizes a rare subset of mature peripheral T cells characterized by the CD3+ CD4- CD8- phenotype. Unlike conventional CAR-T cells, which are typically derived from CD4+ or CD8+ T cells, DNTs exhibit innate-like anti-tumor activity and do not induce graft-versus-host disease (GvHD), facilitating their use as an "off-the-shelf" therapeutic. The platform involves engineering these DNTs to express CARs targeting B-cell antigens such as CD19 and CD22. Clinical and preclinical data suggest that CAR-DNTs are effective in killing leukemia cells, can be cryopreserved without loss of function, and may overcome antigen escape through multi-targeting or "mix-and-match" approaches.
