Overview

CAR-E (CAR-Enhancer) is a novel therapeutic platform developed by researchers at the Dana-Farber Cancer Institute. It is designed to address the limitations of conventional CAR T-cell therapy, particularly the high rate of relapse due to the exhaustion and disappearance of CAR T-cells. The platform works by delivering a fusion molecule that combines a weakened form of interleukin-2 (IL-2) with a tumor antigen (e.g., B-cell maturation antigen (BCMA) for multiple myeloma). This fusion molecule acts externally to enhance the persistence, proliferation, and memory formation of CAR T-cells, thereby extending their activity and preventing cancer relapse. The goal is to enable more durable responses and potentially allow for treatment with fewer CAR T-cells, reducing costs and side effects. Clinical trials are planned to evaluate its safety and optimal dosing.
